Study on Inhibition of Erythropoieis by Estradiol Benzoate


Abstract
Estradiol benzoate was administered to male albino rats in order to elucidate its inhibitory effect on the erythropoiesis, dose response relationship and the time required for maximal inhibition with the following results.
1. Control rats receiving 0.04m1 of vegetable oil daily by intramuscular injection for four weeks exhibited no significant changes in hemoglobin as well as reticulocyte counts before and after experiments.
2. An experimental group, in which daily injections of estradiol benzoate at a dose of 35 ug/kg for four weeks were given intramuscularly, showed decreases in hemoglobin, reticulocyte counts, marrow nucleated red cell counts as well as erythrocyte ¢¥9Fe incorporation.
3. At one, two, three, four and five-day intervals after a single injection of estradiol benzoate at a dose of 100 ,ug/kg, erythrocyte ¢¥9Fe incorporation was measured respectively. The results indicated that maximal inhibition on erythropoiesis occurred at three days
4. A single injection of estradiol benzoate at doses of 10 ;~g/k:g, 25 ,ug/kg, 50 feg/kg and 100 ,ug/kg respectively were given intramuscularly and the erythr~~poietic effect was measured by erythrecyte 59Fe incorporation.
The results disclosed that as the dose of estradiol benzoate increased, the inhibitory effect also proportionally was enhanced.
